Yes you can. If you remember, the Road King Classic still has a 16" spoked wheel. But a 16" wheel from a 2008, '09, '10 and '11 use the same hardware parts (bearings, spacers) so if you get a 16" wheel from any of those years you are good to go.
If the wheel comes with bearings already installed, simply use your old axle. If it doesn't have bearings and spacers, reuse your old spacers but buy new bearings, they are cheap about 30 bucks a piece.
I have a 16", 17" and 18" and I like to change every now and then, lol. My 16" wheel is from a 2008 SG, and I really like the way that fat tire looks.

You can, you just need all the hardware from the newer bike, like the axle, bearings and spacers and the axle hold down nut. The bearings had different dimensions pre-2008

I forgot to mention, I was in the same position as you. I bought my '09 RK with laced wheels, and later on I bought a 16" front wheel on ebay with the intention of sealing them and running WWW tubeless.
When I was about to pull the trigger on the WWW tires, I found out about running a car tire on the back. Now I am not going back to a motorcycle tire, and my laced wheels are sitting in the shed, and I still have the sealant for the spoke holes.
