I've been visiting the Smokies since I was a wee thing. Last year I rode my Heritage up there for the first time and it was a whole new experience.
Everyone seems to be familiar with Deals Gap or the Dragon or highway 129. Its a great ride but do it in the middle of the week if you can. Weekends during the summer are a madhouse. Its popularity is becoming a problem at times.
Cherohala Skyway is a realtivly new road that is truly awsome.
One of my favorites is Little River Road between Townsand and Newfound Gap Road. Its mostly river on side of the road and mostly cliff base on the other. Lush vegetation, moss, ferns, cool air, twisty turns, waterfalls and overlooks all combine to make one heck of nice ride.
Nearby you have the Blue Ridge Parkway which deserves its own weekend.
The locals have learned fast that this area is a motorcycle mecca and many now cater to bikes. I stay in a nice, small motel in Bryson City that is centrally located.
If you live anywhere near the Mid-Atlantic, you need to go. If you dont live near, you need to figure out how to get there and bike anyway

I leave in about 2 weeks to spend 3 nights and 4 days in the area. I'm already excited.
