Route 66 - Best Time?

I don't think that's a concern, considering that the post before you is from over three years ago. 
My first trip along US 66 was the spring of 1959. Things have changed juuuuust a bit since then.
To answer the op's question, the southern US is best in April-May. The wildflowers are in bloom throughout the desert areas, and across the middle of Texas.
It's really spectacular.
