When adjusting the primary chain, you are to adjust it at the tightest spot on the chain.
My question is, does the transmission need to be in first gear? Rotating the rear wheel while
in neutral, primary chain will not rotate. Thanks for the help.

 

You'll have much more sucess if you use fifth gear

If you still have problems, remove the spark plugs.

 

With the plugs removed!
