I rode about 750 miles this weekend and 99% of this was interstate with the cruise set at 80. The ambient temperature was between 32 and 42. MPG was 25 - 29. I'll try it again when it warms up. I wouldn't be surprised if the engine never got to or stayed at normal operating temp...
I think it's very realistic to expect at least 35 MPG on the highway. That equates to 210 miles to the tank and at 180, start looking for a gas station. The fuel gauge and range or DTE are very conservative. I've ridden 20 -30 miles after the DTE changed to LO RNG and only put 5.5 gallons in. I understand that MPG don't matter to the fair weather rider, commuter, bar hopper or occasional weekend road tripper... If that were me, it wouldn't matter either. For now, I'll just enjoy the fruits of my labor and know I better be looking for a gas station when I hit about 125 miles on the tank.

It's not either/or, but and/and. You should get a Target Tune and get your bike dynoed. The Target Tune allows your AFR to be tuned way more accurately. If you don't get the Target Tune module, your engine will still work off of narrowband O₂ sensors, which means that a proper tune will put the AFR outside of the sensors' range.

At 80 MPH your mpg is pretty dang low. Winter blend fuel, and 80 MPH usually nets me 38 mpg on my Road Glide, built motor see signature. On the built King, winter blend and 80 mph usually nets me 42 mpg.
