Most of the time a backfire through the carb indicates a very lean condition. Should be to big of a problem. What type of carb do you have?
Try adjusting your mixture screw more to the rich side and start it up again. But if you get back with us on what type carb you have we can help you out a bit more.
Another thing to check would be that your intake seals are correctly sealing. Sometimes the old rubber bands will rot or the intake manifold will shift enough and create an air gap.

yah take your time and make sure to check everything. Make sure to check that you have no intake leaks by getting the bike started (if you can) and take some WD-40 and spray around the intake seals to see if the RPM's increase. If it does then you likely have an intake leak and need to make sure the seals are completely sealing as need be.
Then if all that checks out richen up the mixture a bit, start it up and start to make sure your timing is correct, then re-adjust your carb as need be.

I went through starter relays till we found the problem. The starter button had a short in it and would start the bike some times and then would die and not refire. After what seemed like a million starter rebuilds and relays bought and swapped found the prob when button wire started smoking. It was causing the trouble all the time. ( Was all new harness from HD and when we found the problem ripped whole harness out and built custom one). 1200 miles since rewire and running strong. Good luck electrical problems stink.
