If you go be sure to ride Rt 6. It's name is Grand Army of the Republic Highway!
Apparently it was the road before Rt 66. We road it a couple years ago. Very nice trip.

Another option depending on far you want to go is Letchworth State Park 35 miles south of Rochester, NY. The picture with my bike was taken there Fall 2009. They have camping and cabins if you are interested.

Nice Area, I spent last Memorial weekend riding there with 20 of my closest friends!
Beer and food very cheap, if you are on a budget

Great area to ride, like some have said, the view is a little limited if you're only going there to see it. Try to set up a nice loop ride, south and west of there is a bunch of roads through the parks/forest lands. I remember some pretty twisty roads on the way up there.

Went there last summer, very scenic. Lots of people from Pa never heard of it. The ride to get there was better though. I've been to Pa three times now and I keep going back, just love riding there. Really enjoyed Route 6 also. Here's a couple pics looking down from the top.

A group of us went last may and stayed a couple nights outside of Wellsboro. We came from NW NJ. The trip was very scenic and the wives had a great time. We took the wagon ride along the river and went to the park. People were friendly, the hotel owner let us put up the bikes overnight on the sidewalk under the overhang due to a rain forcast, which I thought was very nice of him. I think a lot of the local economy relies on biker tourism so they are accomodating. Just remember, it's the ride, not the destination, plenty to do along the way.

The Wellsboro Diner is great, the road is great.

We did the ride twice in the Fall
