Default spoke adjustment

Tunedthe spokes on my RKC for my pre-Sturgis ride and found several "dead" sounding spokes. Tightened all but one, which would not tighten, it just turned like the threads where stripped. I will be putting new tires on real soon and will have the rear tire off. My question is: Can a single spoke be replaced ? What's the fix here?
