Hello All,
I have a troubling question and need help. I have a 1996 fuel injected EVO road king. I have intermittent spark in the front cylinder and no spark in the rear cylinder. Ive changed out the spark plugs, wires, coil and even the cam timing sensor. Nothing has worked.

Run the diagnostics and add a pic of your bike to your signature. Some of us were taught with visual aids.
Do you have the Factory Service Manual? It's invaluable.

Welcome to Evoland.

EFI Evo is single fire, make sure you put a single fire coil on it, so moving on after looking at all the items you replaced, we have a wiring issue. Pull the connector off the coil making sure it is receiving a 12 volt signal at all times even when cranking on 1 of the 3 wires to the coil, you can also use the test light clipped to a positive touching the 2 ground sides of the coil wiring, they will flash when cranking. If the ground side is weak or no flash check the connector under the right side of the engine that your ignition module wiring goes thru, it is known to corrode and may not be getting a good ground to the ignition module that sends a ground signal to the ECM that sends a ground signal to the single fire coil. Pull your seat up, check your battery cables for looseness, check your wire harness where it grounds to the frame for looseness or corrosion something as simple as a loose battery cable does weird crap because the ECM falls below 10 volts at crank and is lost.
