Damage?
Not likely. They only move about at very low speed and so have very little impact. Once you get going at twenty mph or so, the beads have found the light spot and settled in. They are pretty much locked in place by centrifugal force.
As tough as a tire carcass is, I can't imagine that you could go far enough on a tire's tread that you would notice anything in the way of damage or wear inside.
